Fluzone® High-Dose Trivalent is an egg-based, inactivated split-virus influenza vaccine formulated with a higher hemagglutinin (HA) antigen content for enhanced immune response. It is supplied as a sterile, preservative-free, latex-free injectable suspension for intramuscular use.

Fluzone High-Dose is a colorless, opalescent liquid.

High-Dose Formulation Advantage

Fluzone High-Dose is manufactured using the same egg-based split-virus platform as standard Fluzone, with an additional concentration step to increase antigen content.

Key Differentiator:

• Higher HA antigen concentration per strain compared to standard-dose influenza vaccines.

The manufacturing process includes:

• Propagation in embryonated chicken eggs
• Harvest of virus-containing allantoic fluid
• Inactivation with formaldehyde
• Purification using sucrose density gradient centrifugation
• Chemical disruption with Triton® X-100 to produce a split-virus formulation
• Additional concentration after ultrafiltration to increase HA antigen levels
• Separate production of each strain before combination into final trivalent formulation

Antigen Content

Standardized according to United States Public Health Service (USPHS) requirements.

Each 0.5 mL dose contains:

• 180 micrograms (mcg) total hemagglutinin (HA)
  • 60 mcg HA per strain
  • 4x the antigen content of standard-dose influenza vaccines (which contain 15 mcg per strain)
