Fluad® Trivalent is an egg-based, adjuvanted, inactivated influenza vaccine indicated for intramuscular administration. It is supplied as a sterile, milky-white injectable emulsion in preservative-free, single-dose prefilled syringes.


Adjuvanted Formulation for Enhanced Immune Response

Fluad is specifically formulated with MF59C.1 (MF59®) adjuvant, a squalene-based oil-in-water emulsion designed to enhance immune response.

MF59C.1 Adjuvant Components (per 0.5 mL dose):

  • Squalene – 9.75 mg

  • Polysorbate 80 – 1.175 mg

  • Sorbitan trioleate – 1.175 mg

  • Sodium citrate dihydrate – 0.66 mg

  • Citric acid monohydrate – 0.04 mg

  • pH range: 6.9–7.7

The adjuvant works by stimulating a stronger immune response, particularly beneficial in older adults whose immune systems may respond less robustly to standard-dose vaccines.


Manufacturing Process

Fluad is produced using traditional egg-based technology:

  • Virus propagated in embryonated hens’ eggs

  • Harvested and clarified via centrifugation and filtration

  • Inactivated with formaldehyde

  • Purified by zonal centrifugation

  • Surface antigens (hemagglutinin and neuraminidase) extracted using cetyltrimethylammonium bromide (CTAB)

  • Combined with MF59C.1 adjuvant to create the final emulsion


Antigen Content

Standardized according to United States Public Health Service (USPHS) requirements.

Each 0.5 mL dose contains:

  • 45 mcg total hemagglutinin (HA)

    • 15 mcg HA from each of the three recommended influenza strains


Residual Trace Components

Each dose may contain trace amounts of:

  • Neomycin (≤ 0.02 mcg)

  • Kanamycin (≤ 0.03 mcg)

  • Hydrocortisone (≤ 0.005 ng)

  • Egg protein (ovalbumin) (≤ 0.4 mcg)

  • Formaldehyde (≤ 10 mcg)

  • CTAB (≤ 12 mcg)
